چکیده
The extraction of high-quality ribonucleic acid (RNA) from tissues and cells is a key step in many biological assays. Guanidinium thiocyanate-phenol-chloroform (AGPC) widely used efficient method to obtain pure RNA most cells. However, it not with some like sperm because they are resistant chaotropic lysis solutions containing guanidinium thiocyanate such as Buffer RLT+ Trizol. Here, we show that disulfide bonds responsible for the chemical resistance reagents. We while β-mercaptoethanol (βME) can increase RLT+, has no effect Trizol leaves intact. measured reduction 2,2′-dithiodipyridine (DTDP) observed βME pH-dependent activity solutions, suggesting pH limiting factor. identified tris(2-carboxyethyl)phosphine (TCEP) an enhancer AGPC retain reducing even at acidic pH. supplemented TCEP allows complete rapid cells, increasing yield by 100-fold resulting optimal quality reverse transcription polymerase chain reaction. Our findings highlight importance cell various macromolecules bulk single-cell assays, be applied other lysis-resistant vesicles, thereby optimizing amount required starting material animals.

Modified acid guanidinium thiocyanate-phenol-chloroform RNA extraction method which greatly reduces DNA contamination.
The most common and consistently successful methods for isolating intact RNA are modifications of the Guanidinium Thiocyanate method of Chirgwin et al. (1). One widely used modification of this method, developed by Chomczynski and Sacchi (2), involves extraction of the guanidinium thiocyanate homogenate with phenol—chloroform at reduced pH. The Acid Guanidinium Thiocyanate Phenol -Chloroform (A...
متن کاملAn improvement of the single-step method of RNA isolation by acid guanidinium thiocyanate-phenol-chloroform extraction.
A new method of total RNA isolation by a single extraction with an acid guanidinium thiocyanate-phenol-chloroform mixture is described. The method provides a pure preparation of undegraded RNA in high yield and can be completed within 4 h. It is particularly useful for processing large numbers of samples and for isolation of RNA from minute quantities of cells or tissue samples.
متن کاملUsing silica particles to isolate total RNA from plant tissues recalcitrant to extraction in guanidine thiocyanate.
The most commonly used protocol of the RNA isolation, the guanidine thiocyanate method, was unsuitable for recalcitrant plant tissues containing a large amount of storage proteins and secondary metabolites. We demonstrated that RNA could bind to the silica particles, which have been used successfully in DNA isolation from various sources, under a high concentration of NaCl in the presence of et...
متن کاملModification of the acid guanidinium thiocyanate-phenol-chloroform method for nuclear RNA isolation.
The acid guanidinium-phenol-chloroform (AGPC) method has been used extensively for total RNA extraction because of its simplicity and ease (1). In our hands, the AGPC method was found to be very useful for the isolation of intact RNA from several tissues and cell lines (3). On the other hand, when we tried to use the AGPC method to extract nuclear RNA, we were faced with difficulties. The main ...
